Cancer immunotherapy is one of the newest treatments of cancer, highlighted as the ‘breakthrough of the year 2013’ by the journal Science. We have identified a novel pathway that may be used for immunotherapy of cancer. Using high-throughput screening and follow-up experiments, we have identified that this pathway affects the antigen presentation by MHC class I and that it is active in various tumor cell lines. Inhibition of this pathway restores MHC class I function and therefore increases tumor immunogenicity. In this project we aim to test whether this inhibition may serve as a novel immunotherapy in various in vivo tumor and metastasis models. Furthermore, the frequency and further mechanism of this immune escape in patients will be analyzed using primary tumor material. The immune potentiating effects of the pathway inhibitors will also be tested on these primary tumor cells. To obtain primary patient-derived material we closely collaborate with various medical scientists at the AvL, AMC and VUmc. Eventually, this work may provide the rationale for a clinical study using this novel immunotherapy.
Where you will work:
The main focus of the Spaapen lab is to improve current and future immunotherapies. Fundamental discoveries from the lab are translated in collaboration with medical and research experts of the Antoni van Leeuwenhoek hospital (AvL), the Academic Medical Center (AMC) and the VU Medical Center (VUmc) in Amsterdam. The research is embedded in the department of Immunopathology, a multidisciplinary department with a strong background in immunology, molecular biology and high throughput techniques. Sanquin provides a lively, internationally oriented, scientific environment with excellent facilities (flow cytometry, confocal microscopy, mass spectrometry, and sequencing).

- salary and employment conditions are in accordance with CAO Sanquin (Collective Terms of Agreement);
- a temporary appointment for a period of 4 years;
- a 36-hour-week;
- working hours will be determined by mutual agreement;
- reimbursement of travel expenses in accordance with CAO;
- 8,33% year-end;
- 201 vacation hours with full time employment.
