Cancer Immunotherapy is one of the newest treatments of cancer, highlighted as the #breakthrough of the year 2013# by the journal Science. One promising form of immunotherapy involves treatment of patients with in vitro expanded T cells isolated from their own primary tumors (Tumor-Infiltrating Lymphocytes or TIL). During expansion however, T cells mostly differentiate into a short-lived phenotype limiting their long-term therapeutic effectiveness. T cell differentiation is known to be driven by external signals that determine their final fate, but only few of these have been identified. In this project we aim to discover novel signals that can skew T cell differentiation to long-lived phenotypes with stem cell properties. To this end, we will generate a unique human secretome library allowing high-throughput screening to identify novel proteins of interest. The activity of found proteins will subsequently be characterized both biochemically and genetically. Eventually, new differentiation regimens will be validated on T cells derived from primary tumor samples obtained in close collaboration with the Antoni van Leeuwenhoek hospital.
This project is a joint effort from the departments of Immunopathology and Hematopoiesis and the Laboratory for Cell Therapy within the Division Research, in collaboration with medical and research experts from the Antoni van Leeuwenhoek hospital. The research is embedded in the departments of Immunopathology and Hematopoiesis, two multidisciplinary departments with strong backgrounds in immunology, molecular biology and high throughput techniques. Sanquin provides a lively, internationally oriented, scientific environment with excellent facilities (flow cytometry, confocal microscopy, mass spectrometry, and sequencing).
