The AMC is more than just a hospital. It is a world in which patient care, research and education are integrated, and in which people are always central. It is an academic world full of possibilities for talented individuals that are looking for a place where they can excel. Thinking big, daring to dream, cooperating internationally. And never letting yourself be put off by ‘It's not possible' or 'It won't work anyway'. That is the AMC mentality.

This project will focus on the role of the gut and lung microbiota in pneumonia and sepsis. The intestinal microbiota has emerged as a key defense system by local support of mucosal immunity as well as proposed modulatory effects on systemic immunity. Sepsis is the consequence of a deregulated host response to a severe infection. We recently completed human and murine studies that demonstrate a potential protective role of the microbiota in the host response against systemic bacterial infection. Now the aim of this project is to evaluate the mechanistic and functional role of both the gut and lung microbiota in sepsis and develop novel treatment options which will be evaluated in murine models of sepsis. This project will make use of patient samples, a human model in which volunteers are instilled with LPS and murine models of bacterial pneumonia. Experience with microbiota research, CRISPR-Cas9 and functional genomics is preferable. An innovative, pro-active and ‘out-of-the box' way of thinking is of importance in this fast moving research field. The project is sponsored by the Netherlands Organisation of Scientific Research (NWO) by a VIDI grant to the group leader (W. J. Wiersinga).
